Introduction: 

Ceffon-OX Tablet is a fixed dose combination of Cefixime and Ofloxacin tablet which is used in used to treat Typhoid, Infections. bronchitis, pneumonia, urinary tract infections (UTIs), and gonorrhea. In this blend of medicine, Cefixime is a third-generation cephalosporin, while Ofloxacin is a fluoroquinolone antibiotic. CDSCO approved this combination(Cefixime+Ofloxacin) as a new drug on 26.04.2010 for the treatment of patients with typhoid fever and urinary tract infection in adults.

It is a very effective synergy of two medicines that are effective against several types of bacteria where many antibiotics do not found effective.

Understanding Cefixime:

Cefixime is a third-generation cephalosporin category antibiotic that was developed in 1979 and approved for medical use in 1989 by FDA in the United States. It is a broad-spectrum antibiotic that kills bacteria. It is used for several types of infections like;

  • Ear infections
  • Sinus infections
  • Urinary tract infections
  • Gonorrhea
  • Typhoid fever
  • Pneumonia
  • Skin infections

Mechanism of Action of cefixime:

Cefixime breakdown the bacterial cell wall that is responsible for the reproduction of bacteria in the body. This process leads to bacteria dying. It is more resistant to beta-lactamase enzymes than second-generation cephalosporins antibiotics. Safety and Precautions:

Common side effects and how to manage them:

CEFIXIME: Side effects of Cefixime drug reactions include diarrhea, dyspepsia, nausea, and vomiting. Hypersensitivity reactions like skin rashes, urticaria, and in some cases, Stevens-Johnson syndrome have been reported.  There is no specific antidote written for Cefixime overdosage. Gastric lavage may be performed. The dialysis process does not remove Cefixime in significant quantities.

Ofloxacin: In general, fluoroquinolones (Ofloxacin) are well tolerated, with most side effects being mild to moderate. On very rare occasions, serious adverse effects occur. Common side effects include gastrointestinal effects such as nausea, vomiting, and diarrhea, as well as headache and insomnia.

CEFIXIME AND OFLOXACIN TABLET USES: 

CEFFON-OX Cefixime Ofloxacin Tablets work as Cefixime is a cephalosporin antibiotic that treats a wide variety of bacterial infections. Cefixime works by stopping the growth of bacteria in the human body.

This antibiotic treats only bacterial infections like Typhoid in children, It is not given in viral infections (such as the common cold, and flu). Using excessive antibiotics unnecessarily can cause antibiotic resistance to future infections.

CEFIXIME-OFLOXACIN COMBINATION EFFECTIVENESS IN TYPHOID FEVER :

India is reaching fast towards Antimicrobial resistance in the context of typhoid fever. Monotherapy is not found more effective for typhoid fever in India now, then the only practical option is combination therapy. Antibiotic combinations that are approved and available in India for the treatment of typhoid fever are very few, one of them is Cefixime-Ofloxacin. The cefixime-ofloxacin combination is approved by Indian Regulatory Authority In-silico studies have demonstrated a positive rationale for combining these two drugs, while in-vitro studies have substantiated the same by showing a strain-specific synergistic and or additive activity between the two drugs against S. Typhi. Clinical studies in Indian patients have shown huge benefits of using the cefixime-ofloxacin combination in typhoid fever. The first benefit is quick time to defervescence (~3 days), the second is the complete clinical cure of patients in ~7 days only, improving the efficacy in relapse cases. this combination reduced the need for hospitalization for Typhoid fever.

This combination also demonstrates a very good tolerability profile on patients. The Association of Physicians of India Recommends the use of this combination in typhoid fever. So that cefixime-ofloxacin is a safe combination in antimicrobial resistance cases.
